The aim of the lecture is to present basic knowledge on methods of molecular biophysics. The plan of the lecture is as follows: 1. Molecular spectroscopy a) Basics of spectroscopy b) Microwave rotational spectroscopy c) Infrared oscillation spectroscopy d) Raman scattering spectroscopy e) UV-VIS absorption spectroscopy f) UV-VIS emission spectroscopy g) Circular dichroism spectroscopy h) Interaction of magnetic field with molecules i) NMR spectroscopy j) EPR spectroscopy 2. Purification and separation of macromolecules. a) Chromatography b) Electrophoresis 3. Hydrodynamical methods a) Ultracentrifugation b) Viscosimetry 4. Determination of chemical and spatial structures of molecules a) Mass spectrometry b) Crystallization and diffraction measurements of crystals 5. Thermodynamic methods a) Calorimetry b) Osmometry 6. Relaxation methods a) Stopped-flow methods b) Perturbation methods 7. Learning outcomes: |
KNOWLEDGE 1. The student knows the basics of experimental techniques used in molecular biophysics. He can describe and explain the results of experiments using the language of mathematics 2. The student knows the principles of measurement systems used in the field of molecular biophysics 3. He is familiar with the basic use of biophysical methods in the study of biological molecules. SKILLS 1. The student can use the known methods and research tools for planning and analysis simple biophysical experiments. 2. He is able to use the knowledge and methodology of physics (experimental methods) to related scientific disciplines: biochemistry and biology. SOCIAL COMPETENCE 1. The student understands the need for continuous learning. 2. He is able to work in groups, including the interdisciplinary teams. 3. He understands and appreciates the importance of intellectual honesty, he is aware of the ethical problems in the context of research integrity, he understands the role of experiments in the verification of biophysical hypotheses, he knows the role of scientific method in knowledge gathering. The student understands the need for systematic reading of scientific journals, in order to broaden their knowledge, he understands the responsibility and social aspects of the practical application of knowledge. |
Assessment methods and assessment criteria: |
1. Two written tests during course 2. Written exam 3. Oral exam after obtaining 60% of points in total from tests and written exam. The grade of the course will be issued on the basis of grades obtained from final exam (written and oral). A person who did not pass the exam during first session may take the exam in the revision session passing the oral exam, after obtaining 60% of the points from the written exam.
